چکیده

Cells exposed to DNA damaging agents, produce different types of structural changes in the chromosome. Repair of these lesions requires synthesis of new DNA molecules, catalysed by specific DNA polymerases. A putative DNA polymerase has been characterized, for its role in DNA damage repair and radiation resistance in D. radiodurans, a bacterium best known for its extraordinary resistance to  radiation. Transgenic Escherichia coli expressing this protein, showed an improved tolerance to both ionizing and non-ionizing radiations while D. radiodurans cells devoid of this protein become sensitive to DNA damage. Molecular studies suggest, this enzyme’s involvement in base excision repair, a crucial process requires for correcting the DNA damage due to oxidative stress in all living systems.

A Decade of Biochemical and Structural Studies of the DNA Repair Machinery of Deinococcus radiodurans: Major Findings, Functional and Mechanistic Insight and Challenges

The Deinococcus radiodurans bacterium is extremely resistant to ionising radiation and desiccation and can withstand a 200-fold higher radiation dose than most other bacteria with no loss of viability. The mechanisms behind this extreme resistance are not fully understood, but it is clear that several factors contribute to this phenotype. Efficient scavenging of reactive oxygen species and repa...

Identification of a Multiprotein Dna Metabolic Complex from a Radioresistant Bacterium Deinococcus Radiodurans

Deinococcus radiodurans R1 (DEIRA) a member of Deinococcae family can survive ~12 kGy dose of γ radiation which can generate about 200 double strands and 3000 single strand breaks per genome. An efficient DNA strand break repair, contributes maximum to the radioresistance of Deinococcus radiodurans. Distinction in DNA strand break repair from others seems to lie in the mechanism of action of DN...

Whole-genome shotgun optical mapping of Deinococcus radiodurans.

A whole-genome restriction map of Deinococcus radiodurans, a radiation-resistant bacterium able to survive up to 15,000 grays of ionizing radiation, was constructed without using DNA libraries, the polymerase chain reaction, or electrophoresis. Very large, randomly sheared, genomic DNA fragments were used to construct maps from individual DNA molecules that were assembled into two circular over...

Genome of the extremely radiation-resistant bacterium Deinococcus radiodurans viewed from the perspective of comparative genomics.

The bacterium Deinococcus radiodurans shows remarkable resistance to a range of damage caused by ionizing radiation, desiccation, UV radiation, oxidizing agents, and electrophilic mutagens. D. radiodurans is best known for its extreme resistance to ionizing radiation; not only can it grow continuously in the presence of chronic radiation (6 kilorads/h), but also it can survive acute exposures t...

PprA Protein Is Involved in Chromosome Segregation via Its Physical and Functional Interaction with DNA Gyrase in Irradiated Deinococcus radiodurans Bacteria

PprA, a radiation-induced Deinococcus-specific protein, was previously shown to be required for cell survival and accurate chromosome segregation after exposure to ionizing radiation. Here, we used an in vivo approach to determine, by shotgun proteomics, putative PprA partners coimmunoprecipitating with PprA when cells were exposed to gamma rays. Among them, we found the two subunits of DNA gyr...
